The short version of incretin fits in a sentence. The long version — which is the one that helps — is below.
Reviewed 2025-12-05. Anything still debated is marked as such rather than presented as settled.
Acting as a triple agonist, the molecule binds the GLP-1, GIP, and glucagon receptors. GLP-1 activity slows gastric emptying and dampens appetite, while GIP signaling contributes to insulin sensitivity and fat metabolism. Glucagon receptor engagement raises energy expenditure and encourages fat breakdown, although it can also elevate blood glucose. Combining three pathways is intended to yield larger weight reduction than single or dual agonists, and researchers continue to examine how the balance among them shapes tolerability.
Clinical studies have reported notable reductions in body weight among participants. Early trials measured safety and explored several dose levels, and later studies tracked body-weight change over months of treatment. Investigators also monitor effects on glycemic markers, liver fat, and blood lipids. Because the compound is still in development, questions about long-term safety, cardiovascular outcomes, and durability after treatment ends remain open.
The three-receptor design places retatrutide in a distinct category relative to older incretin-based therapies. Single agonists target one receptor, and dual agonists target two. Adding a third target broadens the pharmacological footprint and introduces new trade-offs among efficacy, tolerability, and glucose control. How these trade-offs resolve in large trials is a central focus of current research.
The intended pharmacology combines three signals in one molecule. GLP-1 receptor activation reduces appetite and slows gastric emptying, effects already exploited by approved incretin-based therapies. GIP receptor engagement is associated with improved insulin sensitivity and with direct effects on adipose tissue, although how much it contributes to overall outcomes is still debated. Glucagon receptor agonism raises energy expenditure and supports hepatic lipid handling, a mechanism that also tends to increase glucose output. The triple profile is hypothesized to produce a larger metabolic effect than single or dual agonism, but the relative weight of each receptor in humans is not settled.
Human evidence remains limited to controlled studies. A phase 2 trial in adults with obesity reported large, dose-dependent reductions in body weight over 48 weeks, with gastrointestinal events as the most frequently recorded adverse effect. Phase 3 programs designated TRIUMPH, for obesity, and TRANSCEND, for type 2 diabetes, are intended to confirm efficacy and to characterize safety in larger populations. Related studies are examining conditions such as knee osteoarthritis in people with obesity and metabolic liver disease. Open questions include long-term tolerability, effects on lean mass, and what happens after treatment is stopped.
Retatrutide is an investigational synthetic peptide that acts on three receptor targets at once: glucose-dependent insulinotropic polypeptide, glucagon-like peptide-1, and glucagon. It is developed by Eli Lilly and appears in the literature and in trial registries under the code LY3437943. The molecule belongs to a class of engineered peptides designed to resist rapid breakdown and permit infrequent subcutaneous administration. No regulatory agency has approved it for clinical use, and all available human data come from controlled trials rather than from routine practice.
| Property | Value | Notes |
|---|---|---|
| Molecular weight | Approximately 4,700 Da | Peptide of roughly forty amino acids |
| Receptor targets | GLP-1, GIP, glucagon | Triple agonist class |
| Research code | LY3437943 | Internal development designation |
| Compound class | Incretin mimetic peptide | Synthetic, not a biologic extract |
| Development status | Investigational | No general regulatory approval |
Development has progressed through early- and mid-stage human studies in adults with obesity and with type 2 diabetes. Published phase 2 data reported reductions in body weight and improvements in glycemic markers over the treatment period. No regulatory agency has approved the compound for any indication, and it remains available only within controlled research settings. Whether benefits observed in trials translate into durable outcomes after treatment stops is not yet established.
Retatrutide is an investigational synthetic peptide that acts as an agonist at three distinct G protein-coupled receptors. It combines activity at the glucagon-like peptide-1 receptor, the glucose-dependent insulinotropic polypeptide receptor, and the glucagon receptor within a single molecule. This multi-receptor profile distinguishes it from earlier incretin-based compounds that engage one or two of these pathways. Researchers designed the molecule to test whether simultaneous activation produces greater metabolic effects than single or dual agonism alone.
The peptide backbone is chemically modified to resist rapid enzymatic breakdown in the body. A fatty acid side chain promotes binding to serum albumin, which slows renal clearance and supports an extended circulation time. These modifications allow less frequent administration than would be possible with an unmodified peptide. The precise contribution of glucagon receptor activation to the overall metabolic effect remains an area of active investigation, because glucagon raises glucose while also increasing energy expenditure.
Development has progressed from single- and multiple-ascending-dose studies in healthy volunteers into larger randomized trials in adults with obesity, type 2 diabetes, and fatty liver disease. Early reports describe dose-dependent reductions in body weight and improvements in glycemic markers over treatment periods of several months. Whether the glucagon arm adds tolerability cost without added benefit is still debated. Long-term cardiovascular outcomes, effects after treatment stops, and performance in older or comorbid populations are open questions rather than settled findings. Approval status may change, so the current investigational label should be confirmed against regulatory sources.
Retatrutide is an investigational synthetic peptide developed under the code LY3437943, with a backbone derived from glucose-dependent insulinotropic polypeptide. Several non-proteinogenic residues, including alpha-aminoisobutyric acid, appear in that backbone, and a fatty diacid side chain attached through a linker extends circulation time. The molecule carries roughly thirty-nine amino acid units and a total mass near 4.7 kilodaltons. Administration is by subcutaneous injection once weekly. Published work uses both the name retatrutide and the code LY3437943.
== Applications == Because of the ability of acetalated dextran to degrade more rapidly in low pH environments like the phagolysosome of a macrophage or dendritic cell, it has been used as polymeric micro/nanoparticles. Acetalated dextran was originally developed as a vaccine carrier, but has been used for drug delivery, tissue engineering and infectious disease vaccine delivery. Its unique degradation rates have led to finely tuned release of therapeutic proteins and vaccine elements. Ac-DEX has also been shown the allow proteins to be stored outside the cold chain. Formation of nanoparticles with Ac-DEX can be made through standard methods like emulsion, spray drying and electrospray. Using sonication, inorganic nanoparticles have been embedded into Ac-DEX particles to for a composite material for cancer therapy."Prickly Nanoparticles against Cancer". article. DiCaprio is agnostic. His personal life is the subject of widespread media attention, though he rarely grants interviews and is reluctant to discuss his private life. Since his 40s, DiCaprio has been the focus of various reports detailing his involvement with women aged 25 or younger, and has faced criticism for the age disparity of those relationships. In 1999, DiCaprio met Brazilian model Gisele Bündchen, whom he dated until 2005. He was romantically involved with Israeli model Bar Refaeli from 2005 to 2011. He later dated German fashion model Toni Garrn from 2013 to 2014 and later in 2017. DiCaprio was briefly romantically linked with Barbadian singer Rihanna, with the pair attending Coachella together in 2016. DiCaprio successfully sued French tabloid Oops after it made claims that DiCaprio had gotten Rihanna pregnant. DiCaprio was in a relationship with American model and actress Camila Morrone from 2017 until 2022. He has been dating Italian model Vittoria Ceretti since August 2023. DiCaprio owns houses in Los Angeles and apartments in New York City. In 2009, he bought an island, Blackadore Caye, off mainland Belize—on which he is set to open an environmentally friendly resort—and in 2014, he purchased the original Dinah Shore residence designed by architect Donald Wexler in Palm Springs, California. In 2005, DiCaprio's face was severely injured when model Aretha Wilson hit him over the head with a broken bottle at a Hollywood party. As a result, he required seventeen stitches to his face and neck.
Labial reduction occasionally includes the resection of the clitoral hood when the thickness of its skin interferes with the woman's sexual response or is aesthetically displeasing. The surgical unhooding of the clitoris involves a V–to–Y advancement of the soft tissues, which is achieved by suturing the clitoral hood to the pubic bone in the midline (to avoid the pudendal nerves); thus, uncovering the clitoris further tightens the labia minora.
==== United States ==== In April 2020, The Hill reported that markets selling live animals were still operating in the United States and that animal rights activists were calling for the closure of such markets, in addition to their existing calls to close factory farms. Fresh food markets were common in New York City until refrigeration became commonplace in the 20th century. From the 1990s to 2020, the number of live animal markets in New York City nearly doubled. As of 2020, there are more than 80 markets in New York City that stock live animals and slaughter them on-demand for customers. They are mostly poultry markets located in outer-borough immigrant communities where they are culturally significant and pose low public health risks relative to wildlife markets and other types of exotic markets.

=== Leukocyte defects === Due to the central role of leukocytes in the development and propagation of inflammation, defects in leukocyte functionality often result in a decreased capacity for inflammatory defense with subsequent vulnerability to infection. Dysfunctional leukocytes may be unable to correctly bind to blood vessels due to surface receptor mutations, digest bacteria (Chédiak–Higashi syndrome), or produce microbicides (chronic granulomatous disease). In addition, diseases affecting the bone marrow may result in abnormal or few leukocytes.
Smashburger IP Holder LLC, doing business as Smashburger (stylized SmashBURGER), is an American multinational fast-casual hamburger restaurant chain founded in Denver, Colorado. As of 2022, it has more than 227 corporate and franchise-owned restaurants in 35 U.S. states, the District of Columbia and 2 Canadian provinces. Founded in 2007 by Rick Schaden and Tom Ryan, the chain serves "smashed" burgers using a specialized process of cooking them on a flattop grill at a high heat. This technique originated in the Great Lakes region at pressed-chuck burger restaurants, and has been a staple there for decades. The method sears the burger for flavor. These are then topped with additional ingredients and can be customized. At one time, the chain offered unique burgers in each city where its restaurants were located. The menu also includes chicken, turkey and portobello sandwiches as well as french fries, sweet potato fries, fried pickles and other items. Some locations offer the Udi's gluten-free bun. The restaurant saw rapid growth after its first location opened in 2007 and it added several hundred locations within a few years, although a larger slowdown of the "better burger" industry saw it slow its size and expansion plans. Company leaders initially considered an IPO, but Philippine-based quick-service operator Jollibee Group bought a 40 percent stake in the company in 2015, at which time it was valued at $335 million. As of December 2018, Jollibee owns 100% of Smashburger.

== Diet == Like other blowflies, C. vomitoria colonize animal remains, including humans. While adult C. vomitoria feed on nectar, the larvae feed on corpses, the medium in which they grow. However, it has been shown that feeding on processed substrates (food that are modified for human consumption by increasing shelf life and taste through salting, curing, smoking, etc.) provided much better growth than unprocessed substrates such as raw unmodified liver. Because different substrates drastically affected growth, C. vomitoria is best characterized as a specialist that best utilizes processed substrates (minced meats, for example). Its close relative, Calliphora vicina, is a generalist, being able to utilize mixed substrates with equal growth rates. In the case of overcrowding, C. vomitoria competition results in compensation by increased speed of development, leading to smaller larvae and adults. This has complications in forensics because different parts of the body would grow at different rates. Additionally, it has been shown that the fly larvae are able to colonize even buried remains. Growth rates are similar between surface and buried larvae. Usually, these flies lay their eggs around wounds on fresh corpses shortly after death. Right before the pupal stage, the fly larvae that leaves the carrion can burrow into the soil in order to pupate. Then, adult flies emerge. In decaying carcasses, it was found that Calliphoridae flies dominate, especially C. vomitoria. In both spring and fall, C. vomitoria is the primary species found on carcasses. In some cases, C.

This phenomenon was used in the design of light meters and similar devices. Selenium's semiconductor properties found numerous other applications in electronics. The development of selenium rectifiers began during the early 1930s, and these replaced copper oxide rectifiers because they were more efficient. These lasted in commercial applications until the 1970s, following which they were replaced with less expensive and even more efficient silicon rectifiers. Selenium came to medical notice later because of its toxicity to industrial workers. Selenium was also recognized as an important veterinary toxin, which is seen in animals that have eaten high-selenium plants. In 1954, the first hints of specific biological functions of selenium were discovered in microorganisms by biochemist, Jane Pinsent. It was discovered to be essential for mammalian life in 1957. In the 1970s, it was shown to be present in two independent sets of enzymes. This was followed by the discovery of selenocysteine in proteins. During the 1980s, selenocysteine was shown to be encoded by the codon UGA. The recoding mechanism was worked out first in bacteria and then in mammals (see SECIS element).
=== Synthesis Mechanism === When viral HIV-RNA enters the cell, it is accompanied by a reverse transcriptase, an integrase, and a mature HIV-1 PR. The reverse transcriptase converts viral RNA into DNA, facilitating the integrase's role in incorporating viral genetic information with the host cell DNA. The viral DNA can either remain dormant in the nucleus or be transcribed into mRNA and translated by the host cell into the Gag-Pol polyprotein, which would then be cleaved into individual functional proteins (including a newly synthesized HIV-1 PR) by the mature HIV-1 PR.
The trade particularly picked up after the Viking invasions, with major markets at Chester and Bristol supplied by Danish, Mercian, and Welsh raiding of one another's borderlands. At the time of the Domesday Book, nearly 10% of the English population were slaves. William the Conqueror introduced a law preventing the sale of slaves overseas. According to historian John Gillingham, by 1200 slavery in the British Isles was non-existent. Slavery had never been authorized by statute within England and Wales, and in 1772, in the case Somerset v Stewart, Lord Mansfield declared that it was also unsupported within England by the common law. The slave trade was abolished by the Slave Trade Act 1807, although slavery remained legal in possessions outside Europe until the passage of the Slavery Abolition Act 1833 and the Indian Slavery Act, 1843. However, when England began to have colonies in the Americas, and particularly from the 1640s, African slaves began to make their appearance in England and remained a presence until the eighteenth century. In Scotland, slaves continued to be sold as chattels until late in the eighteenth century (on the second May 1722, an advertisement appeared in the Edinburgh Evening Courant, announcing that a stolen slave had been found, who would be sold to pay expenses, unless claimed within two weeks).
