Acting as a triple agonist, the molecule binds the GLP-1, GIP, and glucagon receptors. GLP-1 activity slows gastric emptying and dampens appetite, while GIP signaling contributes to insulin sensitivity and fat metabolism. Glucagon receptor engagement raises energy expenditure and encourages fat breakdown, although it can also elevate blood glucose. Combining three pathways is intended to yield larger weight reduction than single or dual agonists, and researchers continue to examine how the balance among them shapes tolerability.
Clinical studies have reported notable reductions in body weight among participants. Early trials measured safety and explored several dose levels, and later studies tracked body-weight change over months of treatment. Investigators also monitor effects on glycemic markers, liver fat, and blood lipids. Because the compound is still in development, questions about long-term safety, cardiovascular outcomes, and durability after treatment ends remain open.
Each receptor contributes a different physiological effect. Activation of the GLP-1 receptor slows gastric emptying and reduces appetite signaling in the brain. GIP receptor activity influences insulin secretion and lipid handling, while glucagon receptor stimulation raises energy use and fat oxidation. Combining these pathways is intended to produce weight loss beyond what single- or dual-receptor agonists achieve. Researchers attribute the observed potency to simultaneous engagement of all three targets, though the exact contribution of each receptor to overall effect remains under investigation.
Clinical development has advanced through phase 2 trials in adults with obesity and type 2 diabetes. Reported phase 2 results described substantial average weight reduction over roughly forty-eight weeks of weekly dosing. A phase 3 program is ongoing to confirm efficacy and assess long-term safety. Because the compound has not received regulatory approval, it is not available as a prescription product. Public discussion of retatrutide often conflates trial findings with marketed status, an important distinction when interpreting coverage of the topic.

Clinical development has progressed through phase 2 trials in adults with obesity and type 2 diabetes, with phase 3 programs reported as ongoing. Reported outcomes include reductions in body weight and improvements in glycemic measures over defined treatment periods. Whether these effects translate into durable benefits after treatment ends remains an open question. Long-term safety data across broad populations are not yet complete, and regulatory decisions have not been announced.
Retatrutide is an investigational synthetic peptide engineered to activate three distinct hormone receptors within a single molecule. It targets the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor simultaneously. This triagonist design distinguishes it from earlier incretin-based compounds that act on one or two of these pathways. Structural modifications relative to native gut hormones extend its residence time in circulation. The molecule remains under clinical evaluation and is not approved for any indication.
Receptor activation produces downstream effects that differ by tissue. GLP-1 receptor signaling influences appetite regulation and insulin secretion in a glucose-dependent manner. GIP receptor activity contributes to metabolic handling of nutrients and may modulate adipose tissue. Glucagon receptor engagement raises energy expenditure and promotes hepatic lipid turnover, though the balance among these actions in humans is still being characterized. Preclinical models showed reductions in body weight and improved glycemic markers.
Development has progressed through early- and mid-stage human studies in adults with obesity and with type 2 diabetes. Published phase 2 data reported reductions in body weight and improvements in glycemic markers over the treatment period. No regulatory agency has approved the compound for any indication, and it remains available only within controlled research settings. Whether benefits observed in trials translate into durable outcomes after treatment stops is not yet established.
Retatrutide is an investigational synthetic peptide that acts as an agonist at three distinct G protein-coupled receptors. It combines activity at the glucagon-like peptide-1 receptor, the glucose-dependent insulinotropic polypeptide receptor, and the glucagon receptor within a single molecule. This multi-receptor profile distinguishes it from earlier incretin-based compounds that engage one or two of these pathways. Researchers designed the molecule to test whether simultaneous activation produces greater metabolic effects than single or dual agonism alone.

Red blood cell concentrates are produced either from whole blood or by apheresis. Production from whole blood is far more common than apheresis due to collection and production efficacy as well as economical purposes. When red blood cell concentrates are produced from whole blood, the whole blood is first separated through centrifugation (usually between 3000 to 5000 x g). The red blood cells are denser than plasma and the other present blood cells (platelets, white blood cells) and settle at the bottom of the blood bag. After centrifugation, the red blood cells are separated from the other components (the majority of the plasma, platelets and white blood cells) through the use of an extractor (also referred to as blood press). After extraction, an additive solution is usually added in a ratio of 1:1.5 to 1:2. The purpose of the additive solution is to maintain adequate viscosity, provide nutrients and ATP/GTP building blocks and reduce haemolysis generation throughout blood bank storage. Choice of additive solution has an impact on the red blood cell viability and, thereby, shelf life (expiry date) of the red blood cell concentrate. Usually, shelf life is limited to 4 to 6 weeks, provided that the red blood cell concentrates are stored in adequate conditions (2-6 °C). Commercial additive solutions are typically based on saline. They usually contain glucose, adenine, mannitol and, sometimes, phosphate and guanosine. The additive solution has no, or very little, buffering capacity, but buffering is provided by the red blood cells themselves.
=== Hypopituitarism === An adequate cortisol response is defined as a rise to greater than 550 nmol/L. Patients with impaired cortisol responses (less than 550 but greater than 400 nmol/L) may only need steroid cover for major illnesses or stresses. An adequate GH response occurs with an absolute response exceeding 20 mU/L.
=== Hypoaldosteronism === An ACTH stimulation test for aldosterone can help in determining the cause of hypoaldosteronism, with a low aldosterone response indicating a primary hypoaldosteronism of the adrenals, while a large response indicating a secondary hypoaldosteronism. The most common cause of this condition (and related symptoms) is Addison's disease; it is typically treated by fludrocortisone, which has a much longer persistence (1 day) in the bloodstream.

==== Stability ==== According to Sassen (1997) the role of hydrates at chemosynthetic communities has been greatly underestimated. The biological alteration of frozen gas hydrates was first discovered during the MMS study entitled "Stability and Change in Gulf of Mexico Chemosynthetic Communities". It is hypothesized that the dynamics of hydrate alteration could play a major role as a mechanism for regulation of the release of hydrocarbon gases to fuel biogeochemical processes and could also play a substantial role in community stability. Recorded bottom-water temperature excursions of several degrees in some areas such as the Bush Hill site (4–5 °C at 500-metre (1,600-foot) depth) are believed to result in dissociation of hydrates, resulting in an increase in gas fluxes (MacDonald et al., 1994). Although not as destructive as the volcanism at vent sites of the mid-ocean ridges, the dynamics of shallow hydrate formation and movement will clearly affect sessile animals that form part of the seepage barrier. There is potential of a catastrophic event where an entire layer of shallow hydrate could break free of the bottom and considerably affect local communities of chemosynthetic fauna. At deeper depths (>1,000 metres (3,300 feet)), the bottom-water temperature is colder (by approximately 3 °C) and undergoes less fluctuation. The formation of more stable and probably-deeper hydrates influences the flux of light hydrocarbon gases to the sediment surface, thus influencing the surface morphology and characteristics of chemosynthetic communities.
Some drugs used for weight loss work by increasing energy expenditure. Two of the earliest weight loss drugs, 2,4-dinitrophenol and thyroid hormone, increase cellular energy metabolism, but both were withdrawn from use due to risks. Adrenergic agonists, especially those that work on the beta-2 adrenergic receptor, increase energy expenditure. Although some such as clenbuterol are used without medical approval for weight loss, none have achieved approval for this indication due to cardiac risks. Other drugs such as atypical antipsychotics are believed to reduce energy expenditure.
Fructose level in the semen may be analysed to determine the amount of energy available to the semen for moving. WHO specifies a normal level of 13 μmol per sample. Absence of fructose may indicate a problem with the seminal vesicles. The semen fructose test checks for the presence of fructose in the seminal fluid. Fructose is normally present in the semen, as it is secreted by the seminal vesicles. The absence of fructose indicates ejaculatory duct obstruction or other pathology.
=== Biological applications === Initially developed for geochemical and related research, NanoSIMS is now utilized by a wide variety of fields, including biology and microbiology. In biomedical research, NanoSIMS is also referred to as multi-isotope imaging mass spectrometry (MIMS). The 50 nm resolution allows unprecedented resolution of cellular and sub-cellular features (as reference, the model organism E. coli is typically 1,000 to 2,000 nm in diameter). The high resolution that it offers allows intracellular measurement of accumulations and fluxes of molecules containing various stable isotopes. NanoSIMS can be used for pure cultures, co-cultures, and mixed community samples. The first use of NanoSIMS in biology was by Peteranderl and Lechene in 2004, who used a prototype of NanoSIMS to examine and measure carbon and nitrogen isotopes of eukaryotic cells. This study was the first time that carbon and nitrogen isotope ratios were directly measured at a sub-cellular scale in a biological sample.
=== Pharmacodynamics === NBOMe-LAD is known to act as an agonist of the serotonin 5-HT2A receptor and to interact with other receptors, but shows dramatically reduced potency compared to LSD in vitro. At the serotonin 5-HT2A receptor, it had 37-fold lower affinity, 148-fold lower activational potency in terms of calcium release, and around half the maximal efficacy in terms of calcium release relative to LSD. On the other hand, NBOMe-LAD had only about 4-fold lower potency in terms of β-arrestin recruitment along with similar activational efficacy for this pathway relative to LSD. NBOMe-LAD produces the head-twitch response, a behavioral proxy of psychedelic effects, in rodents, but with greatly reduced potency and maximal efficacy relative to PRO-LAD and analogues. Its ED50Tooltip median effective dose for inducing the head-twitch response was 13-fold lower than that of PRO-LAD and its maximal effect was about one-third that of PRO-LAD. However, the most efficacious dose of NBOMe-LAD was 3.2 mg/kg whereas that of PRO-LAD was 1 mg/kg.

It is a synthetic peptide classified as a triple receptor agonist. It engages the GLP-1, GIP, and glucagon receptors at once. It is investigated for metabolic and weight-related conditions rather than approved for general use.
No regulatory agency has approved it for routine medical use. It remains an investigational compound studied in controlled trials. Access outside research settings is not established.
Each receptor influences energy balance through different pathways. Combining them is intended to add fat loss from lowered intake and higher expenditure. The optimal balance among the three effects is still under study.
Retatrutide is an investigational peptide that activates the GLP-1, GIP, and glucagon receptors at the same time. It is being studied for obesity and type 2 diabetes and has not been approved for clinical use. The internal code LY3437943 refers to the same molecule.
