Retatrutide is an investigational synthetic peptide that acts on three receptor targets at once: glucose-dependent insulinotropic polypeptide, glucagon-like peptide-1, and glucagon. It is developed by Eli Lilly and appears in the literature and in trial registries under the code LY3437943. The molecule belongs to a class of engineered peptides designed to resist rapid breakdown and permit infrequent subcutaneous administration. No regulatory agency has approved it for clinical use, and all available human data come from controlled trials rather than from routine practice.
The intended pharmacology combines three signals in one molecule. GLP-1 receptor activation reduces appetite and slows gastric emptying, effects already exploited by approved incretin-based therapies. GIP receptor engagement is associated with improved insulin sensitivity and with direct effects on adipose tissue, although how much it contributes to overall outcomes is still debated. Glucagon receptor agonism raises energy expenditure and supports hepatic lipid handling, a mechanism that also tends to increase glucose output. The triple profile is hypothesized to produce a larger metabolic effect than single or dual agonism, but the relative weight of each receptor in humans is not settled.

Clinical studies have reported notable reductions in body weight among participants. Early trials measured safety and explored several dose levels, and later studies tracked body-weight change over months of treatment. Investigators also monitor effects on glycemic markers, liver fat, and blood lipids. Because the compound is still in development, questions about long-term safety, cardiovascular outcomes, and durability after treatment ends remain open.
The three-receptor design places retatrutide in a distinct category relative to older incretin-based therapies. Single agonists target one receptor, and dual agonists target two. Adding a third target broadens the pharmacological footprint and introduces new trade-offs among efficacy, tolerability, and glucose control. How these trade-offs resolve in large trials is a central focus of current research.
Retatrutide is an investigational synthetic peptide engineered to activate three distinct hormone receptors within a single molecule. It targets the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor simultaneously. This triagonist design distinguishes it from earlier incretin-based compounds that act on one or two of these pathways. Structural modifications relative to native gut hormones extend its residence time in circulation. The molecule remains under clinical evaluation and is not approved for any indication.
Receptor activation produces downstream effects that differ by tissue. GLP-1 receptor signaling influences appetite regulation and insulin secretion in a glucose-dependent manner. GIP receptor activity contributes to metabolic handling of nutrients and may modulate adipose tissue. Glucagon receptor engagement raises energy expenditure and promotes hepatic lipid turnover, though the balance among these actions in humans is still being characterized. Preclinical models showed reductions in body weight and improved glycemic markers.
Retatrutide is an investigational peptide developed by a pharmaceutical company as a multi-receptor agonist for treating obesity and type 2 diabetes. The compound emerged from research into gut-hormone analogues that act on several receptors simultaneously rather than on a single target. Early preclinical work examined how combined activity at three distinct receptors might produce greater metabolic effects than single-receptor compounds. Published phase 2 results have described substantial reductions in body weight among participants, although the compound remains unapproved in most jurisdictions as of the mid-2020s.
Pharmacologically, retatrutide acts as a triple agonist at the glucagon-like peptide-1 receptor, the glucose-dependent insulinotropic polypeptide receptor, and the glucagon receptor. Activation of the first two receptors is associated with improved insulin secretion and reduced appetite. The glucagon receptor component is thought to increase energy expenditure, a mechanism that distinguishes this molecule from dual-agonist compounds. Researchers continue to investigate how the three activities interact and whether the combined profile offers advantages that justify additional clinical testing.
Several questions about the compound remain unresolved. The durability of weight reduction after treatment stops, the frequency of gastrointestinal side effects, and the long-term cardiovascular profile are topics of ongoing study. Regulatory submissions and phase 3 trial outcomes have not been fully reported in the public literature. Because most available data come from controlled trials rather than general-population use, conclusions about effectiveness outside study settings are provisional. The distinction between established findings and open questions matters when interpreting early coverage of the drug.
== Hepatalin Action == The human body stores nutrient energy that it gets from meals by partitioning it between fats and glycogen. Hepatalin acts selectively on muscle, heart, and kidneys to store nutrient energy as glycogen. Hepatalin does not act on the liver or fat cells (adipocytes) or intestines. Insulin acts mainly on fat cells and the liver, storing fat in fat depots throughout the body, and glycogen and fat in the liver. In a healthy state, the majority of glucose uptake is accounted for by hepatalin action in muscle. In response to an intravenous injection of insulin after a meal, hepatalin action accounted for approximately 55% of the glucose uptake (in rats) and 66% (in humans). The partitioning of the nutrient energy storage process in a healthy body is dependent on the ratio of insulin and hepatalin action. If hepatalin action is decreased, glucose levels after a meal rise higher and for longer, and the pancreas must secrete much more insulin to manage the nutrient processing. Nutrient partitioning shifts from glycogen in muscle to lipids, with elevated blood and organ triglycerides resulting. If hepatalin action is reduced chronically, the metabolic consequences account for the predictable, chronological development of the dysfunctions known to be associated with the metabolic syndrome aka syndrome X. It has been proposed that hepatalin is the missing link in understanding and managing obesity, prediabetes, and type 2 diabetes. Hepatalin action decreases with age, is worsened by a sugar supplemented diet.
== Research == The steroidal antiandrogen cyproterone acetate has been studied for blocking the testosterone flare at the start of buserelin therapy in men with prostate cancer. While cyproterone acetate for two weeks eliminates the biological and biochemical signs of the flare, no benefits on prostate cancer outcomes were observed. Very low doses of buserelin nasal spray have been assessed for increasing testosterone levels and fertility in men with oligoasthenozoospermia and hypogonadotropic hypogonadism.
== History == Artificial ligament research began in the World War I era. In the first documented case of an artificial ligament in 1914, Dr. Corner utilized a piece of silver filament as synthetic graft to reconstruct a ruptured anterior cruciate ligament (ACL). A ligament made of silk was used to replace an ACL in 1918. In the early 1980s, technological progress in chemistry and materials science promoted the development of medically suitable materials. Doctors utilized these synthetic materials in clinical applications. The Food and Drug Administration (FDA) approved an artificial ligament made of Gore-Tex for use in ACL reconstruction in 1986. The design of artificial ligaments in the 1980s consisted of two major parts: a relatively stiff cable or tape, and silicone rubber cylinders on one or both ends. The cable or tape was usually made of polyethylene, nylon or carbon fiber. The silicone rubber cylinder varied in size to fit different sized patients. Theoretically, the flexibility of the silicone rubber would allow some deformation under relatively low loads, and the artificial ligament would stiffen to maintain its shape under higher loads. Practically, this design never achieved its goal to mimic the property of a natural ligament. The mechanical performance of the artificial ligaments was inadequate for widespread clinical application. In the long term, performance loss, complications, and failure occurred. Material deterioration contributed to the ineffectiveness of early artificial ligaments. Issues would occur in the months and years following treatment. J.E.

This is in contrast to many progestins, such as 19-nortestosterone derivatives (e.g., norethisterone, levonorgestrel, dienogest) and 17α-hydroxyprogesterone derivatives (e.g., cyproterone acetate, medroxyprogesterone acetate), which do bind to the AR and have been associated with significant androgenic or antiandrogenic effects depending on the progestin in question. Due to its lack of androgenic and antiandrogenic activity, and hence lack of masculinizing and feminizing effects, progesterone is one of the few progestogens that is suitable for use during pregnancy in women at risk for preterm birth or recurrent miscarriage.
==== Same-sex marriage ==== On June 14, 2004, the Cherokee Nation Tribal Council voted to officially define marriage as a union between a woman and man, thereby excluding same-sex marriage. This decision came in response to an application by a lesbian couple submitted on May 13. On December 9, 2016, same-sex marriage was legalized through an opinion by Todd Hembree, the Cherokee Nation's attorney general. In the opinion, Hembree stated that the 2004 law violated the Cherokee Constitution, which requires equal treatment of tribal citizens. He issued the opinion because the director of the tribe's tax commission sought a decision as to whether the tribe could issue a vehicle tag to a same-sex couple married outside the tribe's jurisdiction.
Evidence of the use of dye in pottery are found in most cases on the upper part of ceramic basins, on the inside surface, the areas in which the reduced dye-solution was exposed to air, and underwent oxidation that turned it purple. In the Late Bronze Age Hala Sultan Tekke on Cyprus was a major exporter of purple-dyed textiles with the dye coming from the mucus gland of the murex sea snail. The production of Murex purple for the Byzantine court came to an abrupt end with the sack of Constantinople in 1204, the critical episode of the Fourth Crusade. David Jacoby concludes that "no Byzantine emperor nor any Latin ruler in former Byzantine territories could muster the financial resources required for the pursuit of murex purple production. On the other hand, murex fishing and dyeing with genuine purple are attested for Egypt in the tenth to 13th centuries." By contrast, Jacoby finds that there are no mentions of purple fishing or dyeing, nor trade in the colorant in any Western source, even in the Frankish Levant. The European West turned instead to kermes dye provided by the insect Kermes vermilio, known as grana, or crimson. In 1909, Harvard anthropologist Zelia Nuttall compiled an intensive comparative study on the historical production of the purple dye produced from the carnivorous murex snail, source of the royal purple dye valued higher than gold in the ancient Near East and ancient Mexico.

== Vapor-ion, charge transfer reaction == The analytes are in the vapor phase. This includes breath, odors, VOCs, and other molecules with low volatility that, due to the constant improvements in sensitivity, are detectable in the vapor phase despite their low vapor pressure. Analyte ions are produced via gas-phase chemical reactions, where charging agents collide with the analyte molecules and transfer their charge. In secondary electro-spray ionization (SESI), a nano-electrospray operated at high temperature produces nanodroplets that evaporate very rapidly to produce ions and protonated water clusters that ionize the vapors of interest. SESI is commonly used for the analysis of trace concentrations of vapors being able to detect low volatility species in the gas phase with molecular masses of up to 700 Da.
== Plot == The Combine have opened an interdimensional portal in place of the destroyed Citadel, to summon reinforcements and defeat the Resistance. Outside City 17, Gordon Freeman and Alyx Vance escape the wreckage of a train they used to flee the city. They proceed to a transmission station, where they make contact with Dr. Isaac Kleiner and Dr. Eli Vance, who have arrived at the White Forest rocket facility. Kleiner and Eli learn that a copied Combine transmission Alyx is carrying may be able to close the portal. Gordon and Alyx reach an abandoned mine, where Alyx is critically wounded by a Combine Hunter. A vortigaunt leads them to an underground outpost, where Gordon is instructed to help gather larvae from an antlion colony to heal Alyx. After the larvae are gathered, vortigaunts begin to heal Alyx. With their abilities diverted, the G-Man can now connect to Gordon. He reveals that he rescued Alyx from Black Mesa during the Resonance Cascade, and that it is imperative she reaches White Forest. The G-Man instructs an unconscious Alyx to tell Eli to "prepare for unforeseen consequences". After Alyx recovers, they reunite with Eli, Kleiner and her pet robot, Dog, at White Forest and are introduced to Dr. Arne Magnusson. The scientists are preparing a rocket which they plan to use with the code to reverse the portal. After Gordon subdues a Combine attack on the facility, Alyx gives Kleiner a message recorded by Judith Mossman, which contains the location of the Borealis, a vanished Aperture Science research vessel.
=== Polymerization === The ring-opening polymerization of butyrolactone gives polybutyrolactone. The resulting reverts to the monomer by thermal cracking. It is claimed that poly(GBL) is competitive with commercial biomaterial poly(4-hydroxybutyrate), or P4HB. It is further claimed that poly(GBL) is cheaper to make than P4HB, although both are bio-derived.

It is an investigational peptide that activates three hormone receptors: GIP, GLP-1 and glucagon. It is being studied mainly for obesity and type 2 diabetes, and it is not approved for any clinical use. Published information comes from controlled trials rather than from general practice.
No. As of the most recent public information it remains investigational in every jurisdiction. Material sold under this name outside trials is a research chemical, not an approved medicine. Current status should always be checked against regulator notices.
Dual agonists act on two receptors, usually GIP and GLP-1. Retatrutide adds glucagon receptor activity, which is associated with increased energy expenditure. Whether that third component adds clinically meaningful benefit over dual agonism remains an open question.
